Better to have poor wis or more endurance? Poor wisdom is quite rare in my experience. If you aren't pushing for second tier weapon skills or a non-mundane class, there is very little reason not to give wisdom the lowest priority. I've found almost all skills can be raised very easily even at low wisdom. The few that are difficult to raise can be trained under the influence of a few hits of spice. Spice can also be used (abused) to lower stats on the short term to help increase your failure rate, letting you further diminish the low wisdom handicap at higher skill levels.

How does spice lower stats? Does taking more spice over time give you spice resistance? Does the same spice stack?

You do get spice resistance on use. Spices, especially unprocessed stuff, have the highs and the lows. First you get the high, then you get a low. Needless to say, during the low, your stats decrease.
Barring certain exceptions. The spice effects stack up to three pinches as far as positive effect goes.
